Progressive supranuclear palsy is a movement disorder with prominent tau neuropathology. Brain diseases with abnormal tau deposits are called tauopathies, the most common of which is Alzheimer's disease. Environmental causes of tauopathies include repetitive head trauma associated with some sports. To identify common genetic variation contributing to risk for tauopathies, we carried out a genome-wide association study of 1,114 individuals with PSP and 3,247 controls followed by a second stage in which we genotyped 1,051 cases and 3,560 controls for the (...) stage 1 SNPs that yielded P ≤ 10-3. We found significant previously unidentified signals associated with PSP risk at STX6, EIF2AK3 and MOBP. We confirmed two independent variants in MAPT affecting risk for PSP, one of which influences MAPT brain expression. The genes implicated encode proteins for vesicle-membrane fusion at the Golgi-endosomal interface, for the endoplasmic reticulum unfolded protein response and for a myelin structural component. © 2011 Nature America, Inc. The collection presents a variety of promising new directions in Royce scholarship from an international group of scholars, including historical reinterpretations, explorations of Royce's ethics of loyalty and religious philosophy, and contemporary applications of his ideas in psychology, the problem of reference, neo-pragmatism, and literary aesthetics.

The Knower Paradox is an element of the class of paradoxes of self-reference. It demonstrates that any theory Ó which (1) extends Robinson arithmetic Q, (2) includes a unary knowledge predicate K, and (3) contains certain elementary epistemic principles involving K is inconsistent. In this paper I present different versions of the Knower Paradox (both in the framework of the first-order arithmetic and in the modal logic). There are several solutions of the paradox. Some of them I discuss in detail, (...) namely solution developed within modal logic, solution proposed by C. A. Anderson and solution proposed by P. Égré. The common defect of these proposals is that they developed a connection between the concepts of knowledge and provability. Finally, I suggest a solution using the basic ideas of the revision theory of definitions. (shrink)

This paper evaluates a recent method proposed by Jeremy Gwiazda for calculating the value of gambles that fail to have expected values in the standard sense. I show that Gwiazda’s method fails to give answers for many gambles that do have standardly defined expected values. However, a slight modification of his method (based on the mathematical notion of the ‘Cauchy principal value’ of an integral), is in fact a proper extension of both his method and the method of ‘weak expectations’. I show that this method gives an appropriate value when the ‘tails’ of the gambles that are eliminated in the truncation are ‘stable’, but that the value is not appropriate when the tails are not stable. I do not attempt to give an argument for the use of this method, but just note that it is more general than Gwiazda’s method, and is mathematically quite natural. (shrink)

We examine the sub-field of philosophy of science using a new method developed in information science, Referenced Publication Years Spectroscopy (RPYS). RPYS allows us to identify peak years in citations in a field, which promises to help scholars identify the key contributions to a field, and revolutionary discoveries in a field. We discovered that philosophy of science, a sub-field in the humanities, differs significantly from other fields examined with this method. Books play a more important role in philosophy of science (...) than in the sciences. Further, Einstein’s famous 1905 papers created a citation peak in the philosophy of science literature. But rather than being a contribution to the philosophy of science, their importance lies in the fact that they are revolutionary contributions to physics with important implications for philosophy of science. We propose that the pragmatic factors that mediate everyday deduction, such as alternative and disabling conditions (e.g. Cummins et al., 1991) and additional requirements (Byrne, 1989) exert their effects on specific inferences because of their perceived relevance to more general principles, which we term SuperPs. Support for this proposal was found first in two causal inference experiments, in which it was shown that specific inferences were mediated by factors that are relevant to a more general principle, while the same inferences (...) were unaffected by factors not relevant to the general principle. These results were extended to deontic inferences in two further experiments. Taken together, these findings show that unstated superordinate principles play a significant role in certain types of reasoning. Questions raised by the findings for the main theoretical approaches are discussed. (shrink)

Some breeding technology applications are claimed to improve animal welfare: this includes potential applications of genomics and genome editing to improve animals’ resistance to environmental stress, to genetically alter features which in current practice are changed invasively, or to reduce animals’ capacity for suffering. Such applications challenge how breeding technologies are evaluated, which paradigmatically proceeds from a welfare perspective. Whether animal welfare will indeed improve may be unanswerable until proposed applications have been developed and tested sufficiently and until agreement is (...) reached on how to conceptualize animal welfare. Moreover, even if breeding technologies do improve animal welfare, they might be objected to on other ethical grounds. Ethical perspectives on earlier animal biotechnologies are relevant for today’s breeding technologies and their proposed applications, but may need reinterpretation. The current paper applies the concept of telos, which previously figured mainly in debates on classical genetic engineering, to genomic selection and genome editing aimed at improving animal welfare. It critiques current accounts of telos and offers an alternative conceptualization that applies to recently proposed applications of breeding technologies. This account rejects both removing the desire to pursue characteristic activities and altering animal bodies in ways that compromise their ability to perform such activities, but conditionally allows increasing robustness against environmental stress. Our account of telos enriches ethical debate on these breeding technology applications by insisting on the connection between the good life, an animal’s constitution, and its activities, thus countering reductive conceptions of welfare. This paper stems from an ethnographic, multidisciplinary study that explored the views and experiences of practitioners and scientists on social, ethical and clinical dilemmas encountered when working in the area of pre-implantation genetic diagnosis for serious genetic disorders. We focus here on staff perceptions and experiences of working with embryos and helping women/couples to make choices that will result in selecting embryos for transfer and disposal of 'affected' embryos, compared to the termination of affected pregnancies following prenatal diagnosis. Analysis and (...) discussion of our data led us to consider the possible advantages of pre-implantation genetic diagnosis and whether a gradualist account of the embryo's and fetus's moral status can account for all of these, particularly since a gradualist account concentrates on the significance of time (developmental stage) and makes no comment as to the significance of place (in vitro, in utero). I draw attention to one of the most important sources of Kuhn’s ideas in Structure of Scientific Revolutions. Contrary to the popular trend of focusing on external factors in explaining Kuhn’s views, factors related to his social milieu or personal experiences, I focus on the influence of the books and articles he was reading and thinking about in the history of science, specifically, sources in the history of chemistry. I argue that there is good reason to think that the history (...) of chemistry had a profound influence on Kuhn’s thinking, and what is remarkable is that this has eluded our attention for so long. I also argue that his interest in the history of chemistry was due to the influence of James B. Conant and Leonard Nash. (shrink)

Objective: Population based cohort studies involving genetic research have been initiated in several countries. However, research published to date provides little information on the willingness of the general population to participate in such studies. Furthermore, there is a need to discover the optimal methods for acquiring fully informed consent from the general population. We therefore examined the results of a population based genetic cohort study to identify the factors affecting the participation rate by members of the general public and also (...) specifically to examine the impact of different consent procedures on the rate of participation by prospective candidates and their subsequent withdrawal rate from the study.Design: Descriptive analyses.Setting and participants: The study evaluated two non-genetic subcohorts comprising 3166 people attending for a health checkup during 2002, and two genetic subcohorts comprising 2195 people who underwent a checkup during 2003.Main outcome measurements: Analysis endpoints were differences in participation rates between the non-genetic and genetic subcohorts, differences between providing non-extensive and extensive preliminary information, and changes in participation status between baseline and at 6 months.Results: Participation rates in the genetic subcohorts were 4·7–9·3% lower than those in the non-genetic subcohorts. The odds ratios of participation in genetic research were between 0·60 and 0·77, and the OR for withdrawal from the research was over 7·70; providing preliminary extensive information about genetic research reduced the withdrawal risks but worsened participation rates .Conclusions: The general population responded sceptically towards genetic research. It is crucial that genetic researchers utilise an informative and educational consent process worthy of public trust. (shrink)

Against Barnett (2012), I argue that the theory I advance in DeRose 2010 is best construed as one on which ‘"were"ed-up’ future-directed conditionals like ‘If the house were not to be painted, it would soon look quite shabby’ are, in ways important to how they function in deliberation, different in literal content from their ‘straightforward’ counterparts like ‘If the house is not painted, it will soon look quite shabby’. I also defend my way of classifying future-directed conditionals against an attack (...) by Barnett by defending a standard (among philosophers) approach to the basic structure of some conditionals. Finally, I counter Barnett’s charge that by using the concept of deliberation in my account of the meaning of future-directed conditionals, I put an implausibly demanding constraint on what it takes to understand such sentences. Habitat templets are graphical-qualitative models which describe the development of life-history strategies in specific environmental conditions. In the context of the previous models of life-history strategies, life-history theorists focused on the density-dependent factors as the factors determining life-history strategies. With the use of habitat templets, the focus is oriented towards the environmental causal factors, considering density-dependent phenomena as by-products of the environmental impact. This implies an important shift in causality as well as in the worldview of life-history theorists: population is (...) not considered as a closed system isolated from the environment. The object of study is the organism-in-its-environment, as a complex multilevel system. This shift has also methodological consequences: Life-history theory combines holistic and reductionistic insights, using a variety of heuristic models. This imposes a new conception of generality as well as of the structure of scientific theories. (shrink)

Back in Ancient India, Shankaracharya postulated a philosophy which is now known as Advaita. According to Advaita philosophy, the ‘jivãtma’ (individual soul) and ‘Brahmãtma’ (universal soul) are one and the same and these are the only ‘real’ things that exist. Everything else is an illusion. To challenge this almost unshakeable viewpoint, I bring to the fore a book authored by a Nobel Laureate. In 1935, Alexis Carrel’s revolutionary book entitled “Man the Unknown” was published. Though controversial in terms of its (...) content, it delineates the unexplored aspects of the human being – the understanding of mental processes and consciousness, which he claims humans have not mastered. Throughout the book, Carrel suggests that there is something that is unknown in the homo sapiens and hence he titles his work “Man the Unknown.” Based on this, I argue that both the esoteric East and the pragmatic West have not uncovered something “hidden” –the unknown factor/element. Substantial evidence exists for this contention in the scientific community. For example, questions such as among identical twins diagnosed with depression, why does one medication cure depression for one twin and have no effect on the other twin? Ceteris paribus, why does therapy work for one client but not for another client with the same disorder? These questions do not have clear, crystallized answers. Perhaps, if such a thing like an unknown element exists, it would pave the way for the postulation of the Grand Theory of the behavioural sciences. (shrink)

The lepidopteral imaginary that animates Nabokov's relationship to translation can be understood in light of Deleuze and Guattari's notions of ‘apparatus of capture’ and ‘rhizomatic becomings’. On the one hand, Nabokov's scientific fascination with butterflies frames his radically literal approach to translation. The scientific dissection of texts and butterflies ‘captures’ an original and singular content as it flutters, reducing it to the homogenised content of a target source. On the other hand, in Speak, Memory's engagement with ‘multiple metamorphoses, familiar to (...) butterflies’, Nabokov as both literary artist and autobiographical personage becomes-butterfly with translation itself, entering into affective intensities with butterflies that include lepidopteral relations of movement and rest, speed and slowness.
